Could you kindly send us the log files at support@red-gate.com?
To locate the log files please navigate to %USERPROFILE%\Local Settings\Application Data\Red Gate\ANTS Performance Profiler 4 .
This log file will help us to investigate further. -
There's no RedGate folder in my LocalSettings\AppData :-(

Ideally you should have log files in that folder.
Which version of IIS are you using?
As you can try specifying unused port with IIS through Ants Profiler Project Wizard.
Also can you try profiling this application with our ANTS Memory Profiler 5 and ANTS Performance Profiler 5 release candidate builds?
