It should be possible to profile on port 80 with ANTS Profiler 4, but you'll need to reconfigure IIS.
In the following help topic, http://www.red-gate.com/supportcenter/Content.aspx?p=ANTS%20Profiler&c=ANTS_Profiler/help/3.1/ap_profiling_web_apps.htm&toc=ANTS_Profiler/help/3.1/toc22729.htm (Profiling ASP.NET Applications), there's a section at the bottom titled "Using the original port for Web applications hosted in IIS 6".
Of the two procedures given, only "Using IIS 5 isolation mode" will work with ANTS 4 at present. (Please note that these procedures relate to ANTS 3, so references to the Profiler Project Wizard don't apply to ANTS 4 -- you can find the relevant settings in the Performance Profiler Settings dialog in ANTS 4. This help topic will of course be updated for the ANTS 4 release.)
The second procedure ("Using the original port") will apply to ANTS 4 once the restriction on the port number is relaxed -- a change that is planned for the final release of ANTS 4.

I'm a Technical Author with the ANTS team, and am responsible for this particular Support Center content.
The version number given at the top of a page (next to the product graphic) will always be that of the last full release -- in this case, ANTS 3.2. If you look a little lower down the page, you'll see "Learning ANTS Profiler - 4.0beta" in grey text above the topic title. This indicates the version of the help you're looking at, and can differ from the full-release number.
It's rare for us to provide help with a beta release, so having a help version higher than the full-release version isn't something you're likely to see elsewhere in the Support Center. However, if you use the table-of-contents links (in the "Learning ANTS Profiler" panel to the right), you should find you're browsing through the 4.0 Beta help just fine.
For the record, it's just web help for this Beta -- the help file (CHM) link still points to version 3 help.
